چکیده

Introduction It is now almost100 years ago that the first victim with severe dementia associated with neuronal cell death was studied by Alois Alzheimer. By that time such cases were very unusual and Alois Alzheimer had the chance to see only two patients throughout his life. Now Alzheimer’s disease (AD) is a very frequent disorder, which affects millions worldwide. Only within Germany, about 1.2 million patients suffer from this devastating disorder. During the last decades a sharp increase of affected persons is observed, which still continues and, according to recent statistical predictions, becomes very dramatic in the near future. This is due to the enormous increase in our life expectancy. Work on the molecular mechanisms of AD from my laboratory and many others now strongly indicate that AD is an age dependent syndrome, which will affect almost all of us as long as we life long enough.

Disease-oriented Genome Networks Diseases of the Nervous System Network: Systematic Gene Identification and Functional Analyses in Common CNS Disorders Project: Identification and Validation of Parkinson Disease Relevant Genes by Functional Genomics in Cell Culture and Animal Models

Introduction PD is characterized by a loss of dopaminergic neurons in the substantia nigra leading to major clinical and pharmacological abnormalities that are representative for this disease (1). The identification of mutations in different PD associated genes has provided insight into the pathogenesis of this disease (2). In the last years it became obvious that hereditary factors as well as ...

Ionotropic Glutamate Receptors and their Role in Neurological Diseases

Glutamate is extensively and relatively uniformly distributed in the central nervous system (CNS) and its effects mediated by two distinct groups of receptors including Ionotropic and metabotropic glutamate receptors. Concentration of glutamate in the nervous system is much higher than in other tissues. Glutamate receptors play an important role in synaptic transmission, neural plasticity and n...

Disease-oriented Genome Networks Diseases of the Nervous System Network: Systematic Gene Identification and Functional Analyses in Common CNS Disorders Project: Identification of Genes Involved in Neuroprotection from Alzheimer Disease

Introduction Alzheimer disease (AD) is a progressive neurodegenerative disorder, characterized by the formation of amyloid peptides (Aβ) and their deposition in the brain as senile plaques (1). The amyloid peptides Aβ40 and Aβ42, and especially their oligomeric aggregates, are believed to play a crucial and early role in the pathogenesis of AD by causing neurotoxicity (2), development of neurof...

P 96: Role of Thrombin in Inflammatory Central Nervous System (CNS) Diseases

Thrombin is a multifunctional enzyme which has key roles in coagulation cascade and inflammatory events. The pro-inflammatory functions of thrombin occur by different mechanisms including increasing mast cell degranulation, up-regulating the expression of cell adhesion molecules (CAMs) and promoting the secretion of inflammatory chemokines and cytokines. Dysregulated signaling functions of thro...
